Pancragen is a lab-made, four-amino-acid peptide (called KEDW) built to target the pancreas — the organ that makes insulin and controls blood sugar. It's part of the same Russian bioregulator peptide family and was designed with hormone-producing (endocrine) tissue in mind (Adv Gerontol 2014; Molecules 2021).

Mechanism under study

Pancragen attaches to a specific spot on DNA found near genes that control pancreas function, and also sticks to histones (the DNA spools mentioned earlier) (Molecules 2021). In pancreas cells grown in a lab, it turned up several genes that help cells become mature, working pancreas cells (Bull Exp Biol Med 2013), and in aging cells it boosted markers linked to cell survival and growth while reducing a cell-aging marker (Adv Gerontol 2012). The overall idea is that it may help pancreas cells work and repair themselves better.

Human evidence summary

This is the standout human data point in this group: in a study of 33 elderly people with type 2 diabetes plus 30 healthy older people, Pancragen lowered blood sugar, insulin levels, and insulin resistance in the diabetic group, while the group that didn't receive it showed no change (Bull Exp Biol Med 2011). Important caveat: this was not a rigorous "gold standard" trial — it wasn't randomly assigned or blinded (meaning people knew who got the treatment), and it wasn't registered in any public trial database, so it's promising but not conclusive.

Preclinical evidence summary

In elderly female monkeys, daily injections into the muscle for 10 days improved how quickly their bodies cleared sugar from the blood and normalized insulin patterns, with some benefits lasting up to 3 weeks after stopping. Researchers compared its effect to a real diabetes drug (glimepiride) and found Pancragen's effect was real but somewhat smaller and slower (Adv Gerontol 2014; Adv Gerontol 2015). This monkey study is a reasonably strong piece of evidence, even though it's still a small, short study.
