Retatrutide

An investigational multi-receptor peptide studied in metabolic and energy-balance research.

Research overview

Retatrutide is an investigational peptide designed to engage multiple metabolic hormone receptors. Research has focused on how combined receptor signaling can influence energy balance, appetite-related signaling, glucose regulation, and body-composition outcomes.

This profile is intentionally educational rather than instructional. Trinity’s Advanced Research Search should be used for the reviewed evidence summary, terminology, source context, and catalog-linked research information.

Why researchers study it

Its scientific interest comes from combining several signaling pathways in one molecule, making it useful for studying how incretin and glucagon-related pathways interact.

Human evidence summary

In a real study of 338 people over 48 weeks, people taking weekly retatrutide lost a huge amount of weight — from about 9% of body weight at the lowest dose up to about 24% at the highest dose, compared to only about 2% for people taking a fake shot (placebo). At the highest doses, somewhere between 60-83% of people lost 15% or more of their starting body weight, versus just 2% on placebo (Jastreboff, NEJM 2023). More studies have also tested it in people with type 2 diabetes and fatty liver disease, with bigger phase 3 trials still ongoing. So far, there's no data yet on long-term heart health outcomes.

Preclinical evidence summary

The existing research profile doesn't include separate animal-study details for retatrutide — the evidence gathered focuses on human trials since the drug moved fairly quickly into human testing.
