PNC-27 is a lab-made peptide built by fusing two pieces together: a small segment of a natural tumor-suppressor protein called p53 (the part that normally binds to another protein, HDM-2/MDM2), attached to a separate 17-amino-acid piece that helps the whole thing stick to and get into cell membranes (Sarafraz-Yazdi et al., Int J Mol Sci 2022). It was specifically designed as an experimental anti-cancer compound.

Mechanism under study

Here's the clever trick: many cancer cells display an unusual amount of a protein called HDM-2 right on their outer surface (membrane), while healthy cells generally don't. PNC-27 is built to specifically dock onto that surface HDM-2. Once several PNC-27 molecules gather at the same spot on a cancer cell's membrane, they poke a hole (pore) straight through it, causing the cell to rapidly die through a messy process called necrosis (different from the cleaner, more controlled cell-death process called apoptosis). Because healthy cells don't display HDM-2 on their surface the same way, they're mostly left alone (Sarafraz-Yazdi et al. 2022; Do et al., PNAS 2010).

Human evidence summary

There are absolutely no human clinical trials of PNC-27. A search of the official clinical trials registry turned up zero registered studies for this compound.

Preclinical evidence summary

This is a notable gap: while PNC-27 has strong lab-dish (in vitro) evidence, the key mechanistic research paper reports no new animal experiment for PNC-27 itself. The animal tumor-shrinking data that sometimes gets attributed to this peptide actually comes from a related but different peptide called PNC-28, not PNC-27 itself. So as of the source data, there is no confirmed live-animal evidence specifically for PNC-27.
