Melanotan II (MT-II or MT-2) is a lab-made, ring-shaped (cyclic) peptide made of 7 amino acids, designed to mimic the natural skin-tanning hormone alpha-MSH but with a stronger, broader effect. It was developed in research labs studying skin pigmentation (PubChem CID 92432; Dorr et al., Life Sci 1996).
Mechanism under study
MT-II works like a master key that fits several different locks (receptors) in the body, not just one. It turns on tanning by activating skin pigment cells, but it also affects brain and spinal cord receptors tied to sexual arousal, yawning, and appetite — meaning it can cause a tan, but also unintended side effects like spontaneous arousal or reduced hunger, because it's hitting more targets than just skin cells (King et al.).
Human evidence summary
A tiny early-stage study with just 3 people showed that repeated small shots caused noticeable tanning, along with nausea, sleepiness, and unwanted erections lasting 1-5 hours (Dorr 1996). A slightly larger study in 20 men with erectile dysfunction found the drug produced erections in 17 of the 20 men and increased sexual desire more often than a placebo (Wessells et al. 2000). Importantly, there have been no larger, later-stage human trials (the kind normally needed before a drug is approved), and no long-term human safety data exists.
Preclinical evidence summary
Studies in rodents support the idea that one of MT-II's target receptors (MC4R) is responsible for triggering erections, though scientists still disagree about how much a second receptor (MC3R) contributes — this part of the science remains unsettled (King et al.).
