Ipamorelin is a lab-made short chain of 5 amino acids (a pentapeptide) originally developed by the drug company Novo Nordisk under the code name NNC 26-0161 (PubChem). It's designed to trick your pituitary gland (a small hormone-control center in your brain) into releasing more of your own natural growth hormone.
Mechanism under study
Imagine your pituitary gland has a button that releases growth hormone. Ipamorelin pushes that button by attaching to a specific 'ghrelin receptor' (the same receptor that responds to the hunger hormone ghrelin) on the pituitary gland and nearby brain cells. What makes Ipamorelin special compared to older similar peptides is that it's picky — it mostly just releases growth hormone without also triggering the release of stress hormones like cortisol or other hormones like prolactin (Raun et al.).
Human evidence summary
The only completed human study is a study in patients recovering from abdominal surgery, testing whether Ipamorelin could help their bowels start working again faster (study). It was given through an IV twice a day for up to a week in 117 patients. The result: the drug was safe and well-tolerated (patients on Ipamorelin actually had fewer side effects than those on placebo), but it did NOT significantly speed up bowel recovery compared to placebo — the trial technically failed to prove it worked for that use (Beck et al.). There are no human trials showing it helps with muscle, body shape, sleep, or anti-aging — those uses are based on personal stories, not real trials.
Preclinical evidence summary
In pigs, Ipamorelin was just as good at releasing growth hormone as an older, less selective peptide (GHRP-6), but without spiking stress hormones like cortisol — this is the key finding behind why it's marketed as 'cleaner' than older growth hormone peptides (study). In a rat study simulating post-surgery bowel paralysis, a single dose helped the rats have their first bowel movement sooner, and repeated dosing increased their food intake and weight gain (study).
